Inhibition of Osteosarcoma metastasis by Insulin-like Growth Factor Binding Protein 5 (IGFBP5)

چکیده
INTRODUCTION Osteosarcoma (OS) is the most common primary malignancy of bone, with a peak incidence in the second decade of life. Although approximately 80% of patients have either macroor micrometastatic disease at the time of diagnosis, only 10-15% of these lesions can be identified with current imaging modalities. With current treatment regimens, which include wide local resection and chemotherapy, the average five year disease-free survival is only 50-65%. Despite significant advances in surgical techniques and chemotherapeutic regimens over the past few decades, there has been minimal improvement in patient survival. Therefore, there is a critical need to identify molecular markers that provide insight into both the pathogenesis and metastasis of OS.

Insulin-like growth factor binding protein 5 (IGFBP5) functions as a tumor suppressor in human melanoma cells
The insulin-like growth factor binding protein 5 (IGFBP5), which is often dysregulated in human cancers, plays a crucial role in carcinogenesis and cancer development. However, the function and underlying mechanism of IGFBP5 in tumor growth and metastasis has been elusive, particularly in malignant human melanoma. Here, we reported that IGFBP5 acts as an important tumor suppressor in melanoma t...
متن کاملIdentification and characterization of novel IGFBP5 interacting protein: evidence IGFBP5-IP is a potential regulator of osteoblast cell proliferation.
Insulin-like growth factor binding protein-5 (IGFBP5) is a multifunctional protein, which acts not only as a traditional binding protein, but also functions as a growth factor independent of IGFs to stimulate bone formation. It has been predicted that the intrinsic growth factor action of IGFBP5 involves binding of IGFBP5 to a putative receptor to induce downstream signaling pathways and/or nuc...
متن کاملInsulin-like growth factor-binding protein 5 (Igfbp5) compromises survival, growth, muscle development, and fertility in mice.
The insulin-like growth factors (IGFs) are essential for development; bioavailable IGF is tightly regulated by six related IGF-binding proteins (IGFBPs). Igfbp5 is the most conserved and is developmentally up-regulated in key lineages and pathologies; in vitro studies suggest that IGFBP-5 functions independently of IGF interaction. Genetic ablation of individual Igfbps has yielded limited pheno...
متن کاملInsulin-like growth factor binding protein 5 enhances survival of LX2 human hepatic stellate cells
BACKGROUND Expression of insulin-like growth factor binding protein 5 (IGFBP5) is strongly induced upon activation of hepatic stellate cells and their transdifferentiation into myofibroblasts in vitro. This was confirmed in vivo in an animal model of liver fibrosis. Since IGFBP5 has been shown to promote fibrosis in other tissues, the aim of this study was to investigate its role in the progres...
متن کاملGenetic screen identifies insulin-like growth factor binding protein 5 as a modulator of tamoxifen resistance in breast cancer.
Tamoxifen resistance is one of the overarching challenges in the treatment of patients with estrogen receptor (ER)-positive breast cancer. Through a genome-wide RNA interference screen to discover genes responsible for tamoxifen resistance in vitro, we identified insulin-like growth factor binding protein 5 (IGFBP5) as a determinant of drug sensitivity. Specific knockdown of IGFBP5 by retrovira...
